The two sides played out a 0-0 draw the first time around and United manager Louis van Gaal will want his side to progress when you consider the quality sides who were eliminated last month.
In early team news, the BBC reports that Michael Carrick and Ashley Young are both unavailable for United, while Jonny Evans is doubtful with a knock.
Those players aside, van Gaal has the rest of his streamlined squad to call upon following some deadline day departures.
Liam Hughes will not feature for Cambridge, with Harrison Dunk also out injured.
In other news, Real Madrid manager Carlo Ancelotti has been speaking about United loanee Javier Hernandez.
The Mexican international has barely featured since temporarily moving to the Bernabeu, playing only 180 minutes of La Liga football and spending the vast majority of his time on the bench.
But despite that, in comments published by ESPN, Ancelotti confirmed the striker will remain at the club for the duration of his loan spell.
“I don’t know how much playing time (he will get), but he has told me he is happy to stay here and work for us,” the Real boss said.
