Vertonghen is out of action until January with injury, forcing Mauricio Pochettino to adjust his line up.
Spurs selected Toby Alderweireld in defence, with Ben Davies a surprise selection alongside him.
It worked, with Tottenham keeping a clean sheet in a 2-0 win.
Vertonghen praised his teammates after the game, and he also acknowledged his contract extension.
Tottenham activated an option to extend Vertonghen’s deal to 2020 this week, BBC Sport reported.
The option gives Spurs more time to try and persuade the Belgian to sign a longer term contract at the club.
Tottemham will face Chelsea in the semi-finals of the Carabao Cup.
